The teams last met in 1999 and the match is part of Chorley's 130th anniversary celebrations.
The fixture will slot between matches against Ross County and Stevenage for players not on the 2013 Tour presented by Aon.
Chorley manager Garry Flitcroft, the former Manchester City midfielder, commented: "This is a fantastic fixture for our fans to look forward to. I'd like to thank Manchester United for agreeing to send a team to play us.
"As you can imagine they get lots of requests for friendlies so it's a major coup for us to be able to put this game on."
Victory Park has a capacity of 3,700 and demand for tickets is expected to be high.
